The 31-year-old's honour was unveiled on Tuesday, when the two squads for Saturday's All Stars clash at Townsville's Queensland Country Bank Stadium was announced.
Walker, who was also captain in 2019 before missing last year's fixture through injury, will start in the halves for Laurie Daley's side alongside Titans' Jamal Fogarty.
Dallin Watene-Zelezniak and Joe Tapine will share the Maori leadership duties for the clash which will kick-off at 7.10pm AEST.
In other news, it was confirmed on Friday that Mudgee's Glen Willow Stadium will remain the official home of the Charity Shield, between Walker's Rabbitohs and the St George Illawarra Dragons, through the 2023 season.
The 2021 edition of the fixture will take place on Saturday, February 27 - marking the fourth time it's been played in Mudgee.
