All 16 teams will take part in the NRL's celebration of Indigenous culture, which includes running a competition for fans to vote for their top Indigenous team of the past 20 years.
Walker, who captained the Indigenous All Stars team in 2019, has been shortlisted for the honour - with the final team to be unveiled in the coming weeks.
The 30-year-old will start at five-eighth for the Rabbitohs against the Dragons on Thursday night at Netstrata Jubilee Stadium - a fixture which will kick-off round 12.
The Dragons' squad will be headlined by Gerringong pair Jackson Ford and Tariq Sims, as well as Shellharbour's Euan Aitken.
Then on Friday, Moruya's Rhys Kennedy and Pambula-born Jack Williams will both start from their respective benches for the Broncos and Sharks match at Suncorp Stadium.
Narooma's Teig Wilton will also start from the bench for the visitors.
Saturday's action will begin with the Roosters, headlined by Kiama's Josh Morris, hosting the Titans - coached by Milton-Ulladulla's Justin Holbrook - at the Sydney Cricket Ground.
The Roosters have announced they'll be without Kiama winger Brett Morris for an indefinite period after he sustained a small transverse process fracture in his lumbar spine during the side's win over the Warriors.
Later that night, Gerringong's Reuben Garrick and his Sea Eagles will battle the Panthers at Lottoland from 7.35pm.
On Sunday, Batemans Bay's Brad Arthur and his Eels travel to ANZ Stadium to face the Bulldogs.
The Indigenous round will wrap up with the Melbourne Storm, featuring Bega's Dale Finucane, going toe to toe with the Knights at Sunshine Coast Stadium - with the latter being coached by former Batemans Bay Tiger Adam O'Brien.
