At the Sydney Cricket Ground, the Blues won the toss and elected to bat first against Victoria.
Openers Gilkes and Daniel Hughes dominated from the outset, scoring 51 from 49 balls and 40 from 47 deliveries respectively - to lay a strong platform for the hosts.
Following this duo's dismissal, all of their teammates also reached double-figures including Hayden Kerr (43), Baxter Holt (41 not out), Moises Henriques (39), Jack Edwards (33), Chris Tremain (27 not out), Kurtis Patterson (22) and Daniel Sams (21) - as the Blues finished their 50 overs at 7-344.
The pick of the bowlers for the Victorians were Matt Short (2-38) and Brody Couch (2-65).
In reply, Victoria, who were without Nowra's Nic Maddinson who is away with the Australia A squad, struggled with the bat.
Only four batsmen reached double-figures, led by Mackenzie Harvey (55) and Jake Fraser-McGurk (45), was the visitors were all out for 170 in the 33rd over.
Tanveer Sangha starred with the white ball for the Blues, taking 4-21 from his 7.2 overs.
Others to claim multiple scalps were Kerr and Adam Zampa, who took 2-16 and 2-44 respectively.
The victory moves the Blues into second on the One-Day Cup ladder, after two matches - with the first being washed out against Victoria at the MCG on Friday, November 12.
Gilkes and his Blues teammates will now shift gears into their Big Bash League seasons, where the former Ulladulla High School student will once again play for the Sydney Thunder - who open their BBL|11 campaign against the Brisbane Heat on December 6.
