Electing to bat first, the Blues made a strong start thanks to openers Daniel Hughes (58) and Gilkes (28).
Then behind a century by Jack Edwards (108 from 122 balls) as well as handy knocks by Olivier Davies (19) and Sean Abbott (14), the hosts reached 8-251 from their 50 overs.
With the white ball, Jason Behrendorff was the pick of the bowlers for Western Australia, taking 3-46, while skipper Mitchell Marsh claimed 2-34.
In reply, starts from Cameron Green (36), Josh Inglis (29), D'Arcy Short (26), Mitchell Marsh (17) and Josh Philippe (14) laid a solid platform for the Warriors.
But they lost their final four wickets for just eight runs, to be all out for 149 in the 41st over.
Abbott starred with the ball, taking 4-23, while Nathan Lyon (3-38) and Mitchell Starc (2-32) also claimed multiple wickets in the win.
Wicketkeeper Gilkes also contributed one catch in the victory.
