The South Sydney playmaker is one of six changes to Laurie Daley's side that tasted defeat in last year's fixture against the Maori Kiwis.
Thirty-one-year-old Walker, who captained the Indigenous side in 2019, will be joined by Titans' Jamal Fogarty in the halves for the clash at Townsville's Queensland Country Bank Stadium on Saturday, February 20.
Rabbitohs teammate Latrell Mitchell will start at fullback and is joined by Eels' Blake Ferguson, Raiders' Jack Wighton, Sharks' Jesse Ramien and Storm's Josh Addo-Carr in the backline.
Sharks prop Andrew Fifita is also back in the squad after missing last year's clash through injury, with his namesake David Fifita included in the back row alongside experienced forward Wade Graham.
North Queensland hooker Reuben Cotter has won the starting hooker spot after Newcastle utility Connor Watson was ruled out as he continues to recover from an Achilles injury - with Dragons' Josh Kerr and Titans' Tyrone Peachey also named in the starting pack.
Watson, along with Adam Elliott and Brenko Lee, withdrew from Indigenous selection, while Dane Gagai was again not considered by either team.
Sea Eagles playmaker Cade Cust has been included on the extended bench alongside fan-favourite Alex Johnston, Warriors duo Joshua Curran and Jamayne Taunoa-Brown, Titans centre Brian Kelly, Bulldogs back-rower Chris Smith and Dragons enforcer Tyrell Fuimaono.
David Kidwell is coach of the Maori Kiwis side, who won the 2020 fixture 30-16, and his squad features Storm's Reimis Smith, Jesse Bromwich, Brandon Smith, Kenneath Bromwich, Nelson Asofa-Solomona and Jahrome Hughes, Raiders' Charnze Nicoll-Klokstad, Bailey Simonsson and Joseph Tapine, Bulldogs' Dallin Watene-Zelezniak and Jeremy Marshall-King, Panthers' Jarome Luai and James Fisher-Harris, Titans' Kevin Proctor and Patrick Herbert, Roosters' Joseph Manu, Sea Eagles' Dylan Walker, Sharks' Briton Nikora, Broncos' Jordan Riki and Rabbitohs' Benji Marshall.
