Tevita Kuridrani, Folau Fainga'a and Joe Powell will miss the Brumbies' clash with the NSW Waratahs at Canberra Stadium on Saturday.
The Brumbies' major squad reshuffle comes against a side set to field the same 23 in successive weeks for the first time this season.
Connal McInerney will start at hooker at the expense of Fainga'a, while Solomone Kata will start at outside centre in place of Kuridrani.
Powell has been rotated out of the squad with young gun Ryan Lonergan to start at scrumhalf, with Nic White set to make his Super Rugby return off the bench.
The Brumbies will enter the Waratahs battle with a vastly different side to the one which went down to Melbourne a fortnight prior.
In come James Slipper [prop], Murray Douglas, Cadeyrn Neville [both second row], Lachlan McCaffrey [flanker], Tom Wright [wing], Lachlan Lonergan and Nic White [both bench].
Out go Fainga'a, Powell, Kuridrani, Nick Frost and Mack Hansen.
Four players who started against the Rebels in round six will move back to the bench against the Waratahs, with those being Scott Sio, Darcy Swain, Rob Valetini and Tom Cusack.
The competition's form winger Wright returns to the starting side after re-signing with the Brumbies this week, joining Tom Banks [fullback] and Andy Muirhead in the back three.
Douglas and Neville will join forces in the second row, while McCaffrey joins Berry's Will Miller and Pete Samu in the back row.
Former Junior Wallabies teammates Lonergan and Bayley Kuenzle will link up in the halves, while Ryan's brother Lachlan Lonergan has forced his way back into the 23.
"It's really exciting to see some individuals who have worked hard and performed well get an opportunity this week," McKellar said.
"We pride ourselves on our depth and having a whole squad mentality here at the Brumbies and we're confident we've picked the best side to get the job done on Saturday night.
"The Waratahs appear to be full of confidence coming into this game and to say we're excited for the challenge would be an understatement."
