Emergency services were called to a Maryborough address about 3.45am, where a woman was found with serious injuries.
She was treated by paramedics but the woman, 52, died at the scene.
Homicide Squad detectives arrested a 39-year-old Maryborough man near the scene just before noon.
Ambulance Victoria has transported the man to Bendigo Hospital. He is in a stable condition.
The investigation remains ongoing.

A woman is dead and a man is on the run after a suspected homicide in Maryborough last night .
Emergency services were called to a Alma Street address about 3.45am on Monday and found the woman, who is yet to be formally identified, with serious injuries and she died shortly after.
Police say a man, believed to be known to the victim, fled the scene before police arrived.
Homicide detectives are now on scene investigate the death.
