Officers patrolling the M5 at Moorebank saw the woman run a red traffic control light at Heathcote Road, and tried to pull her over about 9.45pm on Saturday, police said.
The woman allegedly continued to drive on Nuwarra Road, going 110km/h in a 60km/h zone, and police began a pursuit.
Police said the driver side-swiped three cars and slammed into the back of a silver Hyundai, before mounting the kerb, hitting a van, and stopping in the front yard of a house.
Officers took the 33-year-old out of the car, along with a 14-month-old girl and a three-month-old boy.
The woman was arrested and breath tested, but returned a negative result, police said.
The woman and children were taken to Liverpool Hospital for medical checks.
She has been charged with several offences, including driving unlicensed, exceeding the speed limit by more than 45km/h, negligent driving, and failing to stop following a police pursuit.
The woman has been refused bail and will appear in court on Monday.
