The 22-year-old will join NSW teammates Sean Abbott, Doug Bollinger, Pat Cummins and Shane Watson in the Australian team, for the three-match series, beginning in Adelaide on November 5.
Maddinson made his international debut against India in October last year and is excited by the prospect of representing his country on home soil for the first time.
“It was very much a surprise, but with the Test guys being away, it has opened up the door,” he said.
“I was very shocked when I found out, but if I’m given the opportunity in any of those three games, I will grab it with both hands,” he added.
Maddinson made 34 on debut in Rajkot last year, where he got to see first-hand what it is like playing at international level.
“It all happened pretty quickly, but it was pretty exciting,” he said.
“Any opportunity you get to play for your country is huge, but it will be pretty special to play in a home series for the first time,” he added.
It has been a big year for Maddinson, who was a part of the NSW team who won the Sheffield Shield earlier this year and he had his first taste of the Indian Premier League with Royal Challengers Bangalore.
“I had a few weeks in Dubai for the IPL before I got injured and hopefully they want me back next year,” he said.
“Getting to play with guys like Chris Gayle, AB de Villiers and Virat Kohli, who was my captain, was a great experience and I hope I get another opportunity soon,” he added.
While he is looking forward to the chance to play Twenty20 for Australia, he still has his sights set on the Baggy Green.
“Shield cricket is my number one focus,” he said.
“We start our four-day season soon and I’m really excited to get out there and defend our title.
“A lot of guys might never even get the chance to play in a final, so winning one was pretty special.
“It was definitely one of my career highlights.”
Maddinson is hoping a bit more consistency with the bat will help him be considered for the test Team in the future.
With the World Cup being held in Australia and New Zealand this summer, winning a place in the 50-over team is also in the back of his mind.
“I think it’s hard not to think about, especially with it being in Australia,” he said.
“You never know, if I’m performing well and scoring runs, it is the best way to put my name forward.”
Maddinson said he is not sure what sort of team South Africa will bring to Australia, but is expecting them to be tough.
The T20 squad will assemble in Adelaide on November 1, ahead of the first international on November 5.
Game two is at the MCG on Friday November 7, while Maddinson is hoping he will get to play in front of his home crowd at ANZ Stadium in Sydney for game three on November 9.
